چکیده

Fatty acid binding protein (FADP) is an attractive vaccine candidate and a drug target for controlling fasciolosis caused by Fasciola gigantica in ruminants. The cDNA encoding F. gigantica FADP was amplified by polymerase chain reaction and expressed in Escherichia coli as a recombinant protein using a prokaryotic expression system. The protein was purified to homogeneity by nickel chelating affinity chromatography and subsequently by ion-exchange chromatography. The recombinant protein thus obtained will be assessed for immuno-prophylaxis and for its immunodiagnostic potential in ruminants agaiust F. gigantica.

Fasciola gigantica Fatty Acid Binding Protein (FABP) as a Prophylactic Agent against Schistosoma mansoni Infection in CD1 Mice

Although schistosomicidal drugs and other control measures exist, the advent of an efficacious vaccine remains the most potentially powerful means for controlling this disease. In this study, native fatty acid binding protein (FABP) from Fasciola gigantica was purified from the adult worm's crude extract by saturation with ammonium sulphate followed by separation on DEAE-Sephadex A-50 anion exc...

Native and Recombinant Fatty Acid Binding Protein 3 from Fasciola Hepatica as a Potential Antigen

Lipid-binding proteins are members of widely distributed protein family. They take part in metabolism of different lipophilic ligands and also have a role in physiological activity, metabolism and disposition of essential hydrophobic compounds (fatty acids, phospholipids, eicosanoids, retinoids, etc.) which are important molecules involved in several cellular processes including gene transcript...

Fasciola gigantica of Ruminants: The phylogenetic analysis based on COX1 sequences

BACKGROUND: Fasciola species are parasitic trematode with world wide distribution that infects wild and domesticated herbivores, particularly ruminants. The aim of the present study was to investigate the intra species variations of F. gigantica, from goats and buffalos isolates in two common geographic climates of Iran. METHODS: Fasciola species were collected from goat, buffalo, sheep, and ca...

Effects of feed restriction and dietary fat type on mRNA expression of liver fatty acid-binding protein (L-FABP) in broilers

Background: Liver fatty acid-binding protein (L-FABP) is the main cytosolic binding site for long chain fatty acids in hepatocytes. FABPs enhance the uptake of fatty acids into the cell by increasing their concentration due to decreasing concentration of unbound fatty acids inside the cell. Objectives: The aim of this study was to evaluate the effects of dietary unsaturated to saturated fatty a...

Determination of somatic and excretory-secretory antigens of Fasciola hepatica and Fasciola gigantica using SDS-PAGE

Human fascioliasis due to unknown species and animal fascioliasis caused by both or one of Fasciola spp. are commonly seen in Iran. To compare electrophoretic patterns of somatic and excretory-secretory antigens of F. hepatica and F. gigantica  by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), the adult flukes were collected from infected slaughtered bovine livers. E/S a...
